Transferrin-modified liposomes triggered with ultrasound to treat HeLa cells

چکیده
Abstract Targeted liposomes are designed to target specific receptors overexpressed on the surfaces of cancer cells. This technique ensures site-specific drug delivery reduce undesirable side effects while enhancing efficiency encapsulated therapeutics. Upon reaching tumor site, these can be triggered release their content in a controlled manner using ultrasound (US). In this study, from pegylated calcein-loaded modified with transferrin (Tf) and US was evaluated. Low-frequency at 20-kHz three different power densities (6.2 mW/cm 2 , 9 10 ) found increase calcein release. addition, transferrin-conjugated (Tf-PEG liposomes) were more sonosensitive compared non-targeted (control) liposomes. Calcein uptake by HeLa cells significantly higher Tf-PEG control further enhanced following exposure low-frequency (at 35 kHz). These findings show that targeted have promising potential as safe effective platform.

Gd(III)-DOTA-modified sonosensitive liposomes for ultrasound-triggered release and MR imaging
Ultrasound-sensitive (sonosensitive) liposomes for tumor targeting have been studied in order to increase the antitumor efficacy of drugs and decrease the associated severe side effects. Liposomal contrast agents having Gd(III) are known as a nano-contrast agent system for the efficient and selective delivery of contrast agents into pathological sites. The objective of this study was to prepare...
متن کاملMechanical release from paramagnetic liposomes triggered by low frequency ultrasound
Introduction Non, or minimally, invasive in vivo visualization of drug release triggered by externally-applied physical stimuli, like heating, light or ultrasound (US) is an emerging topic in molecular medicine. As far as US is concerned, most of the published investigations were performed using nanoor micro-bubbles in which the presence of the gas-filled core makes cavitation (and US detection...
متن کاملTargeting Human Dendritic Cells with Lewis X Modified Liposomes
In this short report, we demonstrate that liposomes bearing the Lewis X trisaccharide on the surface (“Awesosomes”) efficiently target human dendritic cells. We chose a glycolipid with Lewis X trisaccharide headgroup to facilitate the targeted liposome uptake via the DC-SIGN internalization pathway. While no uptake of Awesosomes was detected with wild-type human HEK293 cells, HEK293 cells trans...
متن کاملThermosensitive liposomes modified with poly(N-isopropylacrylamide-co-propylacrylic acid) copolymers for triggered release of doxorubicin.
A novel polymer-modified thermosensitive liposome (pTSL) was developed for the delivery of Doxorubicin (DOX) for cancer therapy. Copolymers containing temperature-responsive N-isopropylacrylamide (NIPAAm) and pH-responsive propylacrylic acid (PAA) were synthesized via reversible addition-fragmentation chain transfer (RAFT) polymerization, yielding copolymers with dual pH/temperature-dependent p...
متن کاملRegulation of HeLa cell transferrin receptors.
HeLa cells were found to have a single class of non-interacting receptors specific for transferrin. Both apotransferrin and diferric transferrin competed equally with 125I-diferric transferrin for receptor binding. Transferrin binding was temperature-dependent and reversible. Binding of transferrin to cells exhibited a KD of 27 nM with a maximum binding capacity of 1.8-3.7 x 10(6) molecules/cel...
